Their helmets were believed to be designed to catch nets and keep them from falling onto the limbs, while their armor stopped tridents. On top of this most depictions we have found of the scissor show them fighting retiarius. While others have been found, the main theory persists that they were usually pitted against retiarius or other scissors.

I always thought they trained with their own class, but then fought the others so that they weren't familiar with how to fight them and it made the show more exciting.
From what we have found, they ate a heavy diet of carbs and plant protein mixed with some fish and meats which made them softer but bulky, as stated by some doctors of the time. But they also did heavy exercises such as weight lifting and anaerobic cardio to help their performance. So I would say powerlifting and burpees combined with a heavy diet of protein and carbs. Tons of calcium too, as most gladiator remains we have found contain more calcium than the average person of the time.
Probably the most important aspect would be the amenities they had at the time. Gladiators were usually like prize fighters of the era and had heated bathrooms, rooms, and advanced plumbing for the time. The exceptions to this were the Noxii, Andabata, and prisoner of war gladiators. They had doctors constantly looking after them and ease of access to the best equipment at the time. So a gym membership would go a long way. A good one with a sauna, or a local YMCA.
