Yes, they will. Once you get used to the grip it really doesn't hurt anymore. I do 180kg zercher shrugs and calf raises and, while being exhausting, they don't really hurt.

I don't think heavy lifts nor carries are sufficient for building core strength, and I'd argue that if it gets to that point for someone, then his core is relatively weak (maybe strongman is an exception).
I think bodyweight exercises like hanging leg raises, hanging windshield wipers, dragon flags and the ab roller (and all of their variations) are the best for building core strength.

It seems old school soviet wrestlers used zercher deadlifts and squats and staples of their weight lifting training
I do zercher's once a week for 1 working set 5-8 reps, 2-3 warmup sets, just simple skill work
